Sensitive detection of peptides in Cytochrome C digest using nano LC with micro Pillar Array Columns (µPAC) and Microsaic 4500 MiD Mass Spectrometer

The compact nature of the Microsaic 4500 MiD® MS system and the combination of low pressure requirements and high separation resolution that PharmaFluidics μPAC™ LC columns can offer have made this combination a perfect fit for performing targeted proteomics studies in an environment where technical resources are limited. This application note describes the Nano LC-MS analysis of cytochrome C digest using a PharmaFluidics μPAC™ column under various elution programs. The mass spectrometer used was the Microsaic 4500 MiD® alongside the standard UV detector for comparison.

Characterizing antibody-drug conjugates using µPACTM-MS

The use of micro pillar array columns (μPACTM) for characterizing antibody-drug-conjugates is presented. Kadcyla® (ado-trastuzumab emtansine) and, for comparison, Herceptin® (trastuzumab) tryptic digests were analyzed. The resolving power offered by the μPACTM column allows an in-depth study of conjugation sites and accommodates the separation of isomeric conjugated peptides. In combination with all-ions MS/MS, conjugates peptides can selectively be recognized facilitating data interpretation.

Herceptine® (trastuzumab) originator and candidate biosimilar tryptic digests were analyzed on a 200 cm μPACTM C18 column and peaks eluting were detected by ultraviolet (UV) spectroscopy and high-resolution mass spectrometry (MS). This way, post-translational modifications could be revealed. This makes μPACTM technology attractive to compare different mAb production batches and to compare mAb originator products to biosimilars.

The use of micro Pillar Array Columns (μPACTM) for monoclonal antibody (mAb) peptide mapping is presented. Remicade® (infliximab) originator and candidate biosimilar tryptic digests were analyzed on a 200 cm μPACTM C18 column and peaks eluting were detected by ultraviolet (UV) spectroscopy and high-resolution mass spectrometry (MS). Minute differences such as point mutations in the protein sequence could be detected. μPACTM could thus be of great value in biosimilar characterization.

Micro Pillar Array Columns (µPACTM) Combined with High-Resolution Mass Spectrometry for Skin Ceramide Profiling

Ceramides, a subclass of sphingolipids, are found in all tissues of the human body. The largest diversity is detected in human skin, more specifically in the uppermost layer of the epidermis, i.e. the stratum corneum, were ceramides play a prominent role in the barrier and permeability properties of the skin. Given the huge structural diversity, skin ceramide profiling requires the use of a highly efficient chromatographic step before mass spectrometric measurement. Micro pillar array columns (μPAC) are perfectly suited in that respect. Their unique benefits are illustrated in this application note.

Peptide separation on micro pillar array columns (µPACTM). Part 1: Cytochrome C tryptic digest

Reversed phase cytochrome c tryptic digest separations are routinely employed for the analysis/quality control of HPLC column performance towards proteomics applications. Easily detectable in UV and with few components, this sample proves to be an ideal tool in demonstrating column performance towards more complex digest separations requiring MS detection. The focus of this application note is to illustrate the performance and versatility of a 200 cm μPAC™ column for peptide separations.

Evaluation of Micro-Pillar Array Columns (µPAC) Combined with High Resolution Mass Spectrometry for Lipidomics

Obtaining insight in the structure and concentration of the wealth of lipids present in biological samples is of significant value in many areas, including biomarker discovery or drug discovery and development. However, this requires efficient separation techniques that can handle the varying physicochemical properties and dynamic range of compounds. In the HPLC 2017 Special issue of LCGC Europe, it is demonstrated how µPAC columns in combination with high-resolution mass spectrometry can tackle these challenges, using human blood plasma lipid samples.
